1) Around 5 to 40 bases at the 5‘-end of a sequence are frequently rather difficult to analyze.
We therefore cut the bases at the very 5‘-end and also edit obviously wrong or missing bases downstream of the cut:
for example (positions refer to the edited sequence)

  • position 2             redundant G was deleted
  • positions 22,23   the missing C and A were introduced
  • position 29           the missing A was introduced
  • positions 32,33   the missing C and A were introduced
  • positions 38,39   the missing C and A were introduced

2) The resolution of a sequence gets unreliable towards its 3‘-end. We therefore cut the bases at the very 3‘-end and also edit obviously wrong bases upstream of the cut (for example position 766 of the upper sequence where the T must be replaced by a G).

3) A peak caused by not completely removed dye-terminators results in a wrong sequence: Compare the bases at positions 38 to 40 of the upper and lower sequence
