siRNA

siRNAs (small interfering RNAs) are double-stranded RNA molecules used to specifically silence ("knockdown") target genes upon delivery into cells.
Usually, they are comprised of 21 nucleotides with a 2-base overhang at the 3' end.

Microsynth offers you a broad range of synthesis options for the generation of high-quality siRNA. Synthesized siRNAs undergo the same stringent quality control (MALDI-TOF, PAGE) as every other oligonucleotide produced at Microsynth. Finally, siRNAs are delivered in solution as annealed duplexes (they can also be delivered as two seperate strands upon request ).

New to RNAi? Microsynth would like to support the inexperienced scientist in the setup of the experiment and the design of siRNAs free of charge. Contact the RNA-customer service  for further information.

Microsynth siRNAs. Example of use.

Hela cells were transfected with 2 variants of lamin A/C specific siRNAs. After 72 hrs of incubation, cells were harvested and total extracts were analyzed by Western Blotting using a lamin A/C specific antibody.

Number 1 is a standard siRNA where as number 2 is a serum-stabilized siRNA developed by Microsynth.

Experiment performed by Dr. Stephan Güttinger at the Institute of Biochemistry, ETH Zurich, Switzerland (curtesy of U. Kutay).

Microsynth siRNAs: Example of use II.
HPLC purified siRNA was compared with desalted siRNA. Two different target sequences (a and b) were selected, the corresponding siRNAs produced at Microsynth's facility, and subsequently transfected into human cells. After 48 hours, cell lysates were compared by Western Blot analysis using an HA-specific antibody. In all cases expression of HA-tagged STK Protein was down-regulated, while the target sequence a showed a clearer effect when compared with the non-silenced negative control . Interestingly, HPLC purified siRNA slightly enhanced the degree of silencing for sequence a.

Important Information

  • 0.2 µmol scaled siRNAs: Benefit from an average delivery time of only 3 business days . If ordered before 1 pm, 0.2 uM scaled siRNAs (desalted) leave Microsynth's production facility within 2 business days (!), providing they pass the Quality Control.
  • 0.040 µmol scaled siRNAs: average delivery time 3 business days.
  • Modifications: Modified siRNAs are available upon request.
  • Quality certification: All siRNAs are produced according to our quality management system and assessed by gel electrophoresis and/or MALDI-TOF analysis before shipment.
  • Delivery:  Duplex siRNA are shipped fully deprotected. They are annealed in solution (40 uM) and ready to use.
  • Ordering: Please use our prepared order forms for placement of your order. In case of higher synthesis scales or modified RNA Oligos please inquire for a special offer.
